Determination of Escherichia C

In the experiment, the bacteria- E. coli were placed in two different sets of mediums - Brain-Heart Infusion (BHI) Broth & Nutrient Broth. The mediums were then set in different conditions (i.e. incubated or room temperature). Two methods were then used to determine the generation time of the E. coli: 1) The Direct Method 2) Indirect Method. By measuring the rate of growth of E. coli one can determine which is the best set of conditions for optimal growth of the species E. coli. For the cultivation of E. coli in the room temperature Brain-Heart Infusion (BHI) Broth the generation time that was determined using the Direct Method was 56.26 and by the Indirect method was 80 minutes. For the cultivation of E. coli in the incubated Brain-Heart Infusion (BHI) Broth the generation time that was determined the Direct Method was 23.15 minutes and by the Indirect method was 32 minutes. For the cultivation of E. coli in the incubated Nutrient Broth the generation time that was determined the Direct Method was 33.44 minutes and by the Indirect method was 38 minutes. From these results, it was determined that E. Coli's optimal growth was in the incubated Brain-Heart Infusion (BHI) Broth. The reason for this is that BHI is a high

coli in the room temperature Brain-Heart Infusion (BHI) Broth the generation time that was determined using the Direct Method was 56. coli (Escherichia coli) were used, they are found in the family of bacteria named Enterobacteriaceae, which is informally referred to as the enteric bacteria. After cells are introduced into a new environment, the population goes through several stages of growth. coli in the room temperature Brain-Heart Infusion (BHI) Broth the slope (Figure 1) had the greatest degree of incline followed by the incubated Nutrient Broth (Figure 2)and then room temperature Brain-Heart Infusion (BHI) Broth (Figure 3). coli: 1) The Direct Method- utilizing dilution and spread plates to actually count the colonies produced by living cells 2) Indirect Method- using the Spec 20 to measure turbidity and as a result the proportionate increase in cell accumulation. ly nutritious infusion medium recommended for the cultivation of fastidious organisms (i. Other errors that might have occurred is misuse aseptic techniques and counting error on colonies of plates and for optical densitiy The advantages of using the Direct Method is it to actually count the colonies produced by living cells versus the Indirect Method which uses the Spec 20 to measure turbidity and as a result the proportionate increase in cell accumulation. 0 ml of culture from the flask into the first dilution tube and vortex the tube to mix contents. The flask was then mixed and 4 ml of the culture was aseptically pipet into one of the empty 13 mm test tubes (this is Time Zero for the Spec 20). The reason for having three different sets of growing conditions is to provide us information about the particular species such as to what type of nutrients needed and range of temperature preferred for optimal growth. The uses of doing a growth curve can be applied to the everyday world and give us insight about that strain of bacteria.
