The purpose of this lab was to get used to working with a very valuable tool in the scientific world; the microscope. During which, everyone within the class learned how to operate a microscope. Light microscope Three pieces of thread Prepared microscope slide An "e" from the newspaper Clean microscope slide Transparent metric ruler Coverslip Medicine dropper Container of pond water Scissors How the Microscope Works and Caring for the Microscope1. Read through lab. Become familiar with parts of microscope.2. Use two hands when carrying microscope. Support the base with one hand and the arm with the other. Place the microscope so the arm id towards you. 3. Locate the two sets of lenses on the microscope, one in the eyepiece, and the other in the objective.4. Locate the metal disk, which is called the turret. The turret revolves, which makes switching powers possible. When an objective is lined up directly underneath the eyepiece lens, a click can be heard. This click should be heard every time a slide is being looked at.


Clean and dry the coverslip and slide when finished. Now locate the fine adjustment knob. Count the number of whole millimeter marks to the right edge of the field of view. There was a great difference in thickness as well. On high power it was so close up that the microorganism appeared for a brief second on the screen and then was gone because of the extremely small field of view. Place the slide on the stage of the microscope. Adjust the objectives until the thread and hair can be clearly seen. For example, when the course adjustment knob is turned forward, the stage moves down. It was impossible for the three threads to all be focused at once. The purpose of the objective lens is to have the ability to adjust the magnification of the scope, while the purpose of the eyepiece is to observe the slide. A microscope should also never be moved after it is positioned properly. It is also not so dim so the objects can hardly be seen without straining one's eyes.
